Principle of the Assay::The Human IL-37 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-37 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human IL-37 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-37 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-37 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-37 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Interleukin 37 (IL-37) also named IL1F7, is a protein that in humans is encoded by the IL1F7 gene. The protein encoded by this gene is a member of the interleukin 1 cytokine family. And it is mapped to 2q13. In vitro expression analysis showed that IL-37 was a secretory protein. And IL-37 could play an important role in the link between innate and adaptive immunity and may be useful for tumor immunotherapy. Meanwhile, IL-37 inhibits the production of inflammatory cytokines in peripheral blood mononuclear cells of patients with systemic lupus erythematosus. In addition, IL-37 can emerged as a dual-function cytokine with intra- and extracellular properties for suppressing innate inflammation.
